Enhancing Flow Cytometry & Cell Sorting with Fluorescent Dyes

Background

Flow cytometry is a powerful analytical technique used in biomedical research and clinical diagnostics for identifying and sorting cell populations. Researchers rely on fluorescent dyes conjugated to antibodies or nucleic acid stains to achieve high-precision detection and classification of cells.

Objective

This study evaluates the performance of Home Algae Biotech's fluorescent dyes—including R-PE, APC, and PerCP—in flow cytometry and cell sorting applications, with a focus on sensitivity, photostability, and efficiency.

Methods

Sample Type: Human peripheral blood mononuclear cells (PBMCs)

Fluorescent Dyes Used: R-PE, APC, PerCP-conjugated antibodies targeting CD4, CD8, and CD19

Equipment: Flow cytometer with 488 nm and 640 nm laser excitation

Evaluation Metrics:
✔ Fluorescence intensity & signal-to-noise ratio (SNR)
✔ Photostability over multiple scans
✔ Sorting efficiency & cell viability post-sorting

Results & Findings

1️. Superior Signal Intensity:

       - R-PE and APC provided high quantum yields, enhancing signal clarity in CD4+ and CD8+ T cell populations.

       - PerCP demonstrated large Stokes shift, reducing spectral overlap and improving multi-color panel efficiency.

2️. Enhanced Photostability:

       - APC and PerCP maintained fluorescence intensity over multiple scans, minimizing photobleaching.

       - R-PE was highly sensitive but required dark storage conditions for long-term stability.

3️. Improved Sorting Efficiency:

       - Cells labeled with Home Algae’s dyes showed >98% purity post-sorting.

       - High fluorescence stability ensured low false positives, leading to accurate cell population identification.

4️. Viability & Post-Sorting Recovery:

       >95% cell viability was observed after sorting, confirming minimal cytotoxicity of Home Algae’s dyes.

Conclusion

This study confirms that Home Algae Biotech's fluorescent dyes are highly effective for flow cytometry and cell sorting, offering:
✔ High sensitivity & brightness
✔ Superior photostability
✔ Minimal spectral overlap
✔ Accurate sorting with high viability

🔹 Application Impact: These findings support the use of Home Algae’s fluorescent dyes in cancer research, immunology, and stem cell studies, enabling researchers to perform high-throughput, multi-parameter analysis with precision and reliability.
